dans le cerveau : A\u03b2 pour<br \/>Alzheimer, \u03b1-syn pour Parkinson. Mais il y a plusieurs modes d\u2019agr\u00e9gation : les oligom\u00e8res, les fibres et les plaques amylo\u00efdes. Parmi ces structures, les oligom\u00e8res sont les formes les plus neurotoxiques et celles qui apparaissent le plus pr\u00e9cocement, au cours de la pathog\u00e9n\u00e8se.<\/p><p>Ces oligom\u00e8res cr\u00e9ent des pores dans la membrane des cellules du cerveau. Ces structures sont appel\u00e9es \u00ab\u00a0pores amylo\u00efdes\u00a0\u00bb. Les ions calcium Ca<sup>2+<\/sup> s\u2019engouffrent dans ces pores, d\u00e9clenchant une cascade neurotoxique : stress oxydatif, hyperphosphorylation de la prot\u00e9ine tau, dysfonctionnement synaptique. Ces \u00e9v\u00e9nements provoquent d\u2019importantes perturbations fonctionnelles au niveau des cellules c\u00e9r\u00e9brales atteintes et \u00e0 terme leur d\u00e9g\u00e9n\u00e9rescence et leur Parkinson.&nbsp;&nbsp;<\/p>\n<p>Dans les deux maladies, cette \u00e9tape implique la fixation des prot\u00e9ines amylo\u00efdes \u00e0 la surface des neurones et des astrocytes, au niveau de structures membranaires particuli\u00e8res&nbsp; appel\u00e9es gangliosides.<\/p>\n<p>Nos chercheurs ont \u00e9lucid\u00e9 le code biologique contr\u00f4lant la reconnaissance des gangliosides par les prot\u00e9ines amylo\u00efdes. Ils ont ainsi pu mod\u00e9liser, \u00e0 l\u2019\u00e9chelle atomique, le processus complet de formation des pores amylo\u00efdes.
